Background
Thymocyte-1 antigen (Thy-1), also known as CD90, is a glycophosphatidylinositol (GPI)-anchored cell surface glycoprotein that is a member of the immunoglobulin superfamily. Expression of Thy-1, originally identified as a marker of thymocyte differentiation in mice, has been found to vary between species. In rats, unlike mice, Thy-1 is expressed on bone marrow cells but not on peripheral T-cells. In both mice and rats, Thy-1 is abundantly expressed on thymocytes and neuronal cells and to a lesser extent on fibroblasts, myofibroblasts, vascular endothelial cells, mammary glands and other non-lymphoid tissues. In many tissues the level of Thy-1 expression changes during cell differentiation.
The exact function of Thy-1 has not been fully determined but studies suggest that Thy-1 may be an important regulator of cell-cell and cell-matrix interactions due to its involvement in a range of biological processes including cell growth and differentiation, cell adhesion and migration, apoptotic signalling and tumour suppression.

Thy-1 antibody is a rabbit polyclonal antiserum obtained from rabbits immunised with purified Thy-1 derived from rat brain.
Product
Each vial contains 100µl polyclonal antiserum containing 0.02% sodium azide.

Specificity
Thy-1 antiserum is directed against rat thymocyte-1 antigen (Thy-1) and also reacts with mouse Thy-1.
Applications
Thy-1 is suitable for the detection of rat and mouse Thy-1 by immunohistochemical staining of frozen tissue sections. After drying, the sections can be either left unfixed or fixed with acetone at 4°C for 4 min or 95% ethanol at 4°C for 15 min or 4% formaldehyde in phosphate buffered saline pH 7.4 at 20°C for 15 min.
Also isolated cells can be immunocytochemically stained by this antiserum.
Optimal antibody dilutions for the different applications should be determined by titration. Recommended range 1:50 – 1:100.
Storage
The antibody is shipped at ambient temperature and may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
