Background
Intercellular adhesion molecule 3 (ICAM3) also known as CD50 (Cluster of Differentiation 50), is a type I transmembrane glycoprotein, which normally contain 2-9 immunoglobulin-like C2-type domains, and binds to the leukocyte adhesion LFA-1 protein. This protein is constitutively and abundantly expressed by all leucocytes and may be the most important ligand for LFA-1 in the initiation of the immune response. It functions not only as an adhesion molecule, but also as a potent signalling molecule. ICAM-3 has previously been implicated in apoptotic cell clearance, although its precise role in the clearance process is ill defined.

MA4 is a mouse monoclonal IgG1/k antibody derived by fusion of mouse Ag8.653 cells with spleen cells from a BALB/c mouse immunized with an ICAM-3/HEK transfectant.

Specificity
MA4 recognizes a linear epitope in domains 1 and 2 of ICAM-3. MA4 robustly inhibits apoptotic cell interaction with phagocytes
Applications
MA4 is suitable for immunoblotting, immunocytochemistry, immunohistochemistry on frozen tissues and flow cytometry. Optimal antibody dilution should be determined by titration; recommended range is 1:100 – 1:200 for flow cytometry, and for immunohistochemistry with avidin-biotinylated horseradish peroxidase complex (ABC) as detection reagent, and 1:100 – 1:1000 for immunoblotting applications.
Storage
The antibody is shipped at ambient temperature and may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
