Background
CD51 (Cluster of Differentiation 51), known as vitronectin receptor α chain, or integrin αV is a type I integral membrane glycoprotein. CD51 forms a heterodimer with integrin β1 (CD29), β3 (CD61), β5, β6, or β8. CD51 contains two disulfide-linked subunits of 125 kD and 24 kD. Many extracellular matrix proteins with RGD-motifs are CD51 ligands. In association with its β chains, CD51 binds vitronectin, von Willebrand factor, fibronectin, thrombospondin, osteopontin, fibrinogen, and laminin. CD51, as an adhesion molecule, plays important roles in leukocytes homing and rolling, mediates bone absorption and angiogenesis. CD51 is expressed on endothelial cells, fibroblasts, macrophages, platelets, osteoclasts, neuroblastoma, melanoma, and hepatoma cells.

NKI-M9 is a mouse monoclonal IgG2a antibody derived by fusion of SP2/0 mouse myeloma cells with spleen cells from a Balb/c mouse immunized with cultured melanoma cells derived from a human melanoma metastasis.

Specificity
NKI-M9 is directed anti the CD51 antigen, alpha chain of vitronectin receptor (VNR-a chain) which forms a complex with: CD61 antigen (GP IIIa or b3 chain); b5 Integrin and b1 Integrin. The platelet a-b3 complex functions as a vitronectin receptor and platelet adhesion to vitronectin is inhibited by NKI-M9.
Applications
NKI-M9 is useful for ELISA, immunoprecipitation, flow cytometry and immunohistochemsitry on frozen sections. Optimal antibody dilution should be determined by titration.
Storage
The antibody is shipped at ambient temperature and may be stored at +4°C. For prolonged storage prepare appropriate aliquots and store at or below -20°C. Prior to use, an aliquot is thawed slowly in the dark at ambient temperature, spun down again and used to prepare working dilutions by adding sterile phosphate buffered saline (PBS, pH 7.2). Repeated thawing and freezing should be avoided. Working dilutions should be stored at +4°C, not refrozen, and preferably used the same day. If a slight precipitation occurs upon storage, this should be removed by centrifugation. It will not affect the performance or the concentration of the product.
